Nice to know the inclination of your brother to join the family business.
I would suggest a solid qualification like MBA before he ventures into the family business. After BCom, he should do an MBA in General Management from a reputed Institute like MDI Gurgaon, Amity University, etc.
For learning the family business aspects theoretically, I would suggest a short diploma in entrepreneurship / family business.

He scored well in the 12th Standard and he is much interested to join family business. As you mentioned, he has enrolled for B Com from DU. So you question is what course, along with BCom can benifit him?
I assume that family has established business and your brother will contribute to the business while studying BCom part time.
Well, considering the education context and already established family business, I would recommend your brother to consider short term courses on entrepreneurship/family business from EDII, Ahmedabad or Indian Institute of Management, Ahmedabad, Lucknow, or Kolkata.
I would not recommend to do any other full time courses in business management. Your brother can start contributing to family business and gain some conceptual, managerial guidance through short-term, experiential courses from reputed institutions.
